การเปรียบเทียบฤทธิ์ของสารสกัดหยาบใบสาบเสือ (Chromolaena odorata) และเปลือกทับทิม (Punica granatum) ในการยับยั้งเชื้อ Staphylococcus aureus

บทคัดย่อ
การวิจัยนี้เป็นการศึกษาเชิงทดลองเพื่อเปรียบเทียบประสิทธิภาพของสารสกัดหยาบใบสาบเสือ (Chromolaena odorata) และเปลือกทับทิม (Punica granatum) ในการยับยั้งการเจริญเติบโตของเชื้อแบคทีเรีย Staphylococcus aureus จำนวน 6 เชื้อแยก (Isolates) ที่คัดแยกได้จากรอยโรคผิวหนังอักเสบเป็นหนองในแมวพันธุ์พื้นเมือง 6 ตัว เชื้อแบคทีเรียนี้พบบ่อยทางคลินิกและมักจะลุกลามจนเป็นเหตุให้แมวเสียชีวิตได้
สารสกัดหยาบเตรียมโดยใช้เอทานอล 95% เป็นตัวทำละลาย สกัดภายใต้อุณหภูมิ 35 °C เป็นเวลา 24 ชั่วโมง ส่วนการทดสอบฤทธิ์ต้านแบคทีเรียใช้วิธี Agar disc diffusion method จำนวน 3 ซ้ำ หาค่าความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อแบคทีเรียได้ (Minimum Inhibitory Concentration; MIC) ด้วยวิธี Broth microdilution และความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อแบคทีเรียได้ (Minimum Bactericidal Concentration; MBC)
ด้วยเทคนิค Streak plate ผลที่ได้พบว่า เปลือกทับทิมให้ผลได้ร้อยละ (Percentage yield) สูงสุด เท่ากับ 34.00 (w/w) ของปริมาณวัตถุดิบที่ใช้ ในขณะที่ใบสาบเสือให้ผลได้ร้อยละเท่ากับ 10.38 (w/w) การทดสอบฤทธิ์ต้านแบคทีเรียที่ความเข้มข้น 250 mg/ml และ 500 mg/ml พบว่า สารสกัดทั้งสองชนิดมีฤทธิ์ในการยับยั้งเชื้อ Staphylococcus aureus ได้ โดยที่ความเข้มข้น 500 mg/ml สารสกัดหยาบเปลือกทับทิมและใบสาบเสือให้ขนาดวงใสเฉลี่ยเท่ากับ 14.33±0.91 และ 7.00±0.69 มิลลิเมตร (mm.) ตามลำดับ แต่ต่ำกว่ายาปฏิชีวนะ Gentamicin (18.28±0.75 mm.) อย่างมีนัยสำคัญทางสถิติ [One-way ANOVA; F(2,51) = 951.19, p < .001] เมื่อเปรียบเทียบรายคู่ พบว่า สารสกัดหยาบเปลือกทับทิมมีประสิทธิภาพสูงกว่าสารสกัดหยาบใบสาบเสืออย่างมีนัยสำคัญทางสถิติ (Post hoc Tukey HSD; p < .001) นอกจากนี้ พบความเข้มข้นต่ำสุดของสารสกัดหยาบเปลือกทับทิมมีฤทธิ์ยับยั้งและฆ่าเชื้อได้สูงที่สุด โดยมีค่า MIC เท่ากับ 3.90 mg/ml และค่า MBC เท่ากับ 7.81 mg/ml ส่วนสารสกัดหยาบใบสาบเสือมีค่า MIC เท่ากับ 7.81 mg/ml และค่า MBC เท่ากับ 62.50 mg/ml จากผลวิจัยสรุปได้ว่าสารสกัดหยาบเปลือกทับทิมมีศักยภาพสูงในการยับยั้งและฆ่าเชื้อ Staphylococcus aureus ดังนั้นสามารถนำไปพัฒนาเป็นทางเลือกสำหรับรักษาโรคผิวหนังอักเสบเป็นหนองในแมวได้
